Monitoring and effect of a tetracycline-fluoranthene combination on subclinical myocardial inflammation and collagen turnover in rainbow trout. The objective of this study was to evaluate the ability of an antibiotic-fluoranthene combination to inhibit bone-associated collagen turnover and to inhibit the production of key markers of inflammation in heart tissue of rainbow trout (Oncorhynchus mykiss). Fish were exposed to 0.1 mg/L triclosan + 2 mg/L fluoroanthene for 7 d. Heart tissue was collected prior to exposure and at seven time points following exposure (days 1, 3, 7, 11, 14, 17, 21 and 24). Inhibition of the activity of the master enzyme in bone collagen synthesis, collagen prolyl hydroxylase, was assessed by measuring the production of 6-hydroxyproline as a surrogate for bone collagen turnover in excised fish. The effects of the antibiotics on the production of key cytokines (interleukin 1β and tumour necrosis factor-α) in heart tissue were also evaluated. The combination of the antibiotics caused a rapid (within a week) inhibition of bone collagen synthesis and increased the production of interleukin 1β and tumour necrosis factor-α in heart tissue. The effect of the antibiotics on the activity of collagen prolyl hydroxylase was partially reversible by day 21. No significant effects of the antibiotics were observed on pro-inflammatory cytokines in heart tissue at any time-point. This study indicates that a combination of tetracycline and fluoroanthene is effective at inhibiting bone-associated collagen synthesis and pro-inflammatory cytokine production, while not affecting the inflammatory status of the heart.
